چکیده
Regulatory T cells (Tregs) participate in the negative regulation of inflammatory reactions by suppressing effector cells. In a number autoimmune disorders, suppressive function and/or Tregs is compromised. The lack active functioning can be restored with adoptive transfer expanded ex vivo autologous Tregs. our study, we traced differentiation and maturation CD4+CD25+FoxP3+CD127low over 7 days cultivation from initial CD4+ under conditions. resulting cell population (eTregs) demonstrated immune profile an increased capacity to suppress proliferation target expression FoxP3 gene was upregulated within time expansion associated gradual demethylation promotor region cell-specific region. Real-time RT-PCR analysis revealed changes genes involved cycle regulation. addition FOXP3, displayed elevated mRNA levels Ikaros zinc finger transcription factors main telomerase catalytic subunit hTERT. Alternative splicing FoxP3, hTERT IKZF family members eTreg maturation. Our data indicate that eTregs develop Treg-specific phenotype functional activity. We suggest are not just but transformed enhanced capacities suppression. findings may influence further development immunosuppressive therapy based on regulatory

Induction of T Regulatory Subsets from Naïve CD4+ T Cells after Exposure to Breast Cancer Adipose Derived Stem Cells
Background: Adipose derived stem cells (ASCs) provoke the accumulation and expansion of regulatory T cells, leading to the modulation of immune responses in tumor microenvironment. Objective: To assess the effect of tumoral ASCs on the trend of regulatory T cells differentiation. Methods: Peripheral blood naïve CD4+ T cells were co-cultured with ASCs derived from breast cancer or normal breast ...
متن کاملT Helper Cells Profile and CD4+CD25+Foxp3+Regulatory T Cells in Polycystic Ovary Syndrome
Background: Polycystic ovary syndrome (PCOS) is considered as the most common cause of female infertility that affects 4-10% of women in the reproductive age. Previous studies have shown the role of a balanced immune response in a successful pregnancy and fertility. Objective: To investigate the T helper cells type 1 (Th1) /Th2/Th17/Treg paradigms in peripheral blood of infertile PCOS compared ...
متن کاملEx Vivo Isolation and Characterization of Cd4+Cd25+ T Cells with Regulatory Properties from Human Blood
It has been known for years that rodents harbor a unique population of CD4(+)CD25(+) "professional" regulatory/suppressor T cells that is crucial for the prevention of spontaneous autoimmune diseases. Here we demonstrate that CD4(+)CD25(+)CD45RO(+) T cells (mean 6% of CD4(+) T cells) are present in the blood of adult healthy volunteers. In contrast to previous reports, these CD4(+)CD25(+) T cel...
متن کاملEx Vivo Generation of Human Alloantigen-Specific Regulatory T Cells from CD4posCD25high T Cells for Immunotherapy
BACKGROUND Regulatory T cell (Treg) based immunotherapy is a potential treatment for several immune disorders. By now, this approach proved successful in preclinical animal transplantation and auto-immunity models. In these models the success of Treg based immunotherapy crucially depends on the antigen-specificity of the infused Treg population. For the human setting, information is lacking on ...
